From Makati CBD take Nagtahan - Quezon Ave - UP I guess, its the shortest route.
From Ayala go to South Avenue turning to Vito Cruz then SSHway then Espana etc.

this may not be the shortest,but it's the safest for me:
from the ninoy aquino statue in ayala,take paseo de roxas, left to makati ave,then to makati mandaluyong bridge, turn right to san francisco (ba yun?) then take nueve de pebrero going to wilson street in greenhills. turn left to ortigas then right to santolan (boni serrano). go staight until you reach p.tuazon ave. in cubao. go straight then turn left to JP Rizal, then right to aurora blvd then left to katipunan ave. go straight until you reach UP

Hmm...most the of routes are already listed.
Take note that if you decide to use C5 and Katipunan, be ready for a lot of fast moving vehicles. although it's quite safe because there are less PUV's that will cut you.
If ever you use Makati-Mandaluyong -> greenhills or shaw to get to C5, get ready for a lot of climbing.
The route through The Fort will offer you less climbs but I think it's a bit longer.
You can also use Buendia then climb the flyover but go down in Kalayaan/C5 BYPASS The Fort.

Sir Ed, the shortest distance between two points is a straight line, if you want to go c-5 then you have to go waaaay around ala - letter - C since C-5 is a circumferencial road it will take you double the distance I guess. But who cares when your on a bike its not about the destination right? ;D

try my not so daily commute route. Makati Ave ---> Makati - Mandaluyong bridge ---> 9 de Pebrero - --> cross Shaw ---> P. Guevarra ----> Wilson ----> Ortigas towards N. Domingo ----> cross Aurora Blvd. ----> Betty Go Belmonte ---> cross E Rodriguez----> Sct. Ybardaloza ----> Kamuning ----> GMA7 (this is where I stop)...from here you can take cross EDSA Malakas St. ---> Kalayaan Ave (via V.Luna Rd) ---> Claret ---> CP Garcia ----> ta dah UP Diliman.
I hope I didnt confuse you but I would be willing to ride with you any time next year after 5PM when I go home. Hope it helped, at a leisurely pace Makati - GMA is 48 minutes (12km from my office to my mom's place)

Yukon has the safest route......
BUT
be sure not to use this route everyday if you have an expensive bike. well, it's a precaution actually coz some secondary roads are not safe enough from bikenappers/ mobster especially in 9 de pebrero (no pun intended) a lot of squatters in that area........ so happy urban assault to you.

that stretch of nueve is scary specially when its dark. good thing i am out this december when the sun sets real early and lots of kids playing those PVC bazookas which shocks/jerks me.

Thanks for the loads of info guys. Made me think twice about the "safety factor" of certain routes. I'll try to use all the routes above and see whatever suits me best. Will update this thread soon ("route review" ika nga).

(this is i think the shortest). from makati cbd, go to fort bonifacio, -> C5 -> katipunan -> UP hmmm.. im not edededed but i can answer the Q depends if you feel like it -- hehehe.. sometimes them bike commuters would throw out a challenge kase kaya ayun, napapa hataw sa ortigas-c5 flyover. my bad.. but pasig-c5 flyover is much more faster than pasig intersection.. be careful sa patintero with the fast moving vehicles though.. and dont forget to use hand signals..

My son is intending to bike from Makati CBD to Ateneo this summer. The route I was thinking of is ayala ave crossing edsa (btw are bikes allowed on edsa?) to the Fort, exiting at Market-market to C-5, then turn right just before the flyover going to katipunan, to the road beside marikina river going towards riverbanks then left and up marcos hi-way (not sure of name) then right to Katipunan then Ateneo. I figured it was an easier climb than having to climb the flyover to katipunan also a little safer as the flyover is only two lanes and the vehicles tend to speed climbing up. Of course he'll have to contend w/ the jeepneys coming from marcos hi-way.

the easiest route from makati cbd to up-diliman is from ayala avenue, cross edsa going to mckinely road, passing forbes park to the fort. inside the fort from mckinely, ride straight, until you turn left to c-5. from c-5, ride straight to the katipunan interchange, passing through the new sm, tiendesitas, and libis. upon reaching katipunan, it's either you turn left in front of miriam or go straight to balara. curahee!
